My idea would be more of an idealisation than a ...practical gameplay mechanic, and it would definitely make the game far harder to make. The idea of certain NPCs being plot locked would work very well, while still keeping some of the origional idea. Certainly more realistic to design.
That said, it depends if the game is story based or more 'sandboxy'- if the only objectives are to get your wife and get out, it would be fairly easy- certain NPCs can help, or they'll try to kill you, chosen at random. A more story driven game would make this impossible.

Alternately, the necromancer that revived you could just be out for revenge, or be reviving you as a power grab against the necromancer conglomerate. I prefer the idea of having shades of grey, instead of a clearly defined good or evil. Seems that a vampire protagonist would be an antihero at best, or at worst a vilain protagonist, and if you run with that, it makes sense that the group that uses such an individual wouldn't be doing it for the best of reasons.

Those seem like a decent set of stats. Anymore run the risk of losing focus, IMO.

I think maybe instead of having 'combos' leading to meteors etc., you could copy magicka. That is to say, lightning means a chained lightning attack medium, earth creates elemental rocks to throw, and certain key combinations could be spells or even new elements (fire+water=steam, fire+ice=water, etc. etc)
That way people could find their own unique, effective combinations without you having to cod them in (For example, in Magicka an extremely powerful spell is steam+lightningX2+arcane (basically lazers), which wasn't specifically coded to do what it does, but is nonetheless the most powerful beam in the game.)

Also, I would have it so that anyone can die- kind of like the STALKER games, where half the time you're taking mission objectives from your friend's dead body. So if the player is looking for his wife, he could find the wife infected, secretly infected or normal- chosen at random. Indeed, I'd have this with all NPCs- for example, there are two or three people who may give you the same map, but some or all of them might turn up dead, infected or secretly infected. Also, mission objectives should not be vital if they're intermediate- for example, it should be possible to shoot everyone from 100m and still save your wife and escape. It would just be bloody difficult.
